dc.description.resumo | Staphylococcus aureus é comumente encontrado em infecções de feridas onde este patógeno compromete o reparo da pele. A lectina isolada das folhas de Schinus terebinthifolius (denominada SteLL) possui ação antimicrobiana e antivirulência contra S. aureus. O estudo avaliou os efeitos da administração tópica de SteLL em feridas de camundongos infectados por S. aureus. No total de 72 camundongos C57/BL6 (6-8 semanas de idade) foram alocados em quatro grupos: (i) feridas não infectadas;(ii) feridas infectadas em não tratadas; (iii) feridas infectadas e tratadas com 32 µg/mL de solução de SteLL; (iv) feridas infectadas tratadas com solução SteLL 64 µg/mL. Uma lesão de aproximadamente 8 mm de diâmetro foi induzida na região dorsal de cada animal e infectadas por S. aureus 432170 (4,0 x 106 UFC/ferida). O tratamento diário começou 1 dia pós-infecção (dpi). A aplicação tópica de ambas as concentrações de SteLL observou-se uma diminuição da severidade da infecção por S. aureus, resultados estes que se correlacionam com a diminuição da carga bacteriana no leito da ferida, reduzindo significativamente a cicatrização de feridas infectadas até o 7º dpi, quando comparada a lesões infectadas não tratadas (reduções de 1,95-4,55 vezes e 1,79-2,90 vezes para SteLL a 32 µg/mL e 64 µg/mL, respectivamente). O tratamento baseado em SteLL também alterou a gravidade da infecção da ferida e reduziu a carga bacteriana (12 vezes para 72 vezes para 32 µg/mL; e 14 vezes para 282 vezes para 64 µg/mL). As feridas tratadas com SteLL mostraram maior deposição de colágeno e restauração da estrutura da pele se comparada aos demais grupos. A carga bacteriana e os níveis de marcadores inflamatórios (IL-6, MCP-1, TNF- α e VEGF) também foram reduzidos por ambas as concentrações de SteLL. Esses resultados corroboram com as propriedades anti-infecciosas relatadas de SteLL, tornando esta lectina uma candidata líder para o desenvolvimento de agentes alternativos para o tratamento de lesões cutâneas infectadas por S. aureus. | por |
dc.description.abstract | Staphylococcus aureus is commonly found in wound infections where this pathogen compromises skin repair. The lectin isolated from Schinus terebinthifolius leaves (called SteLL) has antimicrobial and antivirulence action against S. aureus. The study evaluated the effects of topical administration of SteLL on wounds of mice infected with S. aureus. A total of 72 C57/BL6 mice (6-8 weeks old) were allocated into four groups: (i) uninfected wounds; (ii) infected wounds treated with 32 µg/ml SteLL solution; (iv) infected wounds treated with 64 µg/mL SteLL solution. A lesion of approximately 8 mm in diameter was induced in the dorsal region of each animal and infected by S. aureus 432170 (4.0 x 106 CFU/wound). Daily treatment started 1 day post-infection (dpi). Topical application of both SteLL concentrations showed a decrease in the severity of S. aureus infection, results that correlate with the decrease in bacterial load in the wound bed, significantly reducing the healing of infected wounds up to 7thdpi, when compared to untreated infected lesions (1.95-4.55-fold and 1.79-2.90-fold reductionsfor SteLL at 32 µg/mL and 64 µg/mL, respectively). SteLL-based treatment also altered the severity of wound infection and reduced bacterial load (12-fold to 72-fold for 32 µg/mL; and 14-fold to 282-fold for 64 µg/mL). Wounds treated with SteLL showed greater collagen deposition and restoration of skin structure compared to the other groups. Bacterial load and levels of inflammatory markers (IL-6, MCP-1, TNF-α and VEGF) were also reduced by both SteLL concentrations. These results corroborate the reported anti-infective properties of SteLL,making this lectin a leading candidate for the development of alternative agents for the treatment of S. aureus infected skin lesions. | eng |
